Soreness after unfamiliar exercise typically appears the following morning and peaks a day or two after that. The delay is informative, because it rules out most of the explanations people assume.
Lactic acid is not the cause
Lactate accumulates during hard effort and produces the burning sensation felt at the time. It is cleared from the blood within an hour of stopping.
Anything still present two days later cannot be lactate, and measurements confirm that levels have long since returned to baseline.
The persistence of the lactic acid explanation reflects how well it matches the sensation during exercise rather than any evidence about the days afterwards.
The damage is mechanical and localised
Delayed soreness follows movements where muscle lengthens under load, such as lowering a weight or running downhill, far more than shortening contractions.
Lengthening under tension disrupts the microscopic structures within muscle fibres, producing small-scale damage concentrated in the fibres least prepared for that pattern.
This is why an activity that is not especially tiring can produce severe soreness, while an exhausting session of familiar work produces almost none.
The delay comes from the repair response
Damaged tissue triggers an inflammatory response that recruits immune cells to clear debris and initiate rebuilding. That process takes time to develop.
Swelling within the muscle raises pressure on nerve endings, and inflammatory mediators lower the threshold at which those nerves fire.
The pain therefore tracks the repair process rather than the damage. Its timing reflects when the response peaks, which is typically a day or two after the session.
Soreness is a poor measure of a session's value
Damage is a consequence of novelty more than of effectiveness. A well-trained person doing productive work in a familiar pattern will often feel nothing.
Conversely, severe soreness after a first session mainly indicates that the movement was unfamiliar, which says little about whether the stimulus was appropriate.
Using soreness to judge whether training is working therefore rewards constant variation over progression, which is the opposite of what drives adaptation.
The protective effect after one exposure
Repeating the same activity within a few weeks produces markedly less soreness, an effect that appears after a single prior exposure.
The protection comes partly from structural reinforcement within the fibres and partly from a dampened inflammatory response to the same insult.
Severe soreness accompanied by dark urine, marked swelling or weakness that does not settle is a different matter and requires prompt medical assessment.
